### Consensus consistency validation — 9 October 2026

Consensus browser Pro and connector discovery and fetched paper records were reconciled with independently retrieved original PubMed primary abstracts/status records. The exact query and three fetched records are retained with the consistency audit. This supplementary check used the prior grade visibly; it is not a blind re-score and is not covered by any separately verified adversarial approval of this supplement.

- PASADENA, PMID 35921451, DOI 10.1056/NEJMoa2202867: original Results confirm neither dose improved the global primary outcome or dopamine-transporter imaging meaningfully. This agrees with E51/[51]. Lack of benefit does not establish removal of the intracellular toxic species or disprove every alpha-synuclein mechanism.
- SPARK, PMID 35921450, DOI 10.1056/NEJMoa2203395: original Results confirm no clinical or imaging advantage and stopping for lack of efficacy. This agrees with E55/[55]; it is a separate trial from PASADENA.
- PASADENA extension, PMID 39379705, DOI 10.1038/s41591-024-03270-6: the original abstract describes a favorable exploratory comparison against external PPMI participants. This agrees with E53/[53]. It remains part of NCT03100149, not independent randomized confirmation; publication-type metadata does not make the extension contrast randomized.

**Assessment after reconciliation:** Γ / Moderate held, with SC1 Ζ and SC2–SC4 Γ. All three sources were already in the 65-report inventory; the supplementary check adds zero primary reports or independent causal lines. Source access for this check is abstract/status depth, not a complete-paper audit. The unchanged human-scope blocker is selective toxic-species mediation and generalization, rather than simply counting negative trials.

The supplementary Pro thread is preserved verbatim: [Alpha-synuclein Parkinson causality](https://consensus.app/search/alpha-synuclein-parkinson-causality/HQ86Wa25Rl6VsUDvHSlliw/). Provider syntheses and full-text badges do not establish source access by this reviewer; the matched original-source checks above determine credit.

This dated supplement adds source reconciliation to the original review. Its original narrative, registered scope, source inventory and grades remain historical records. A dated adversarial approval record exists for the separately identified alpha-synuclein.review.md (2026-10-06; see edition-approval-audit.json). Its scope is that recorded edition. This supplement has no separate approval and no coverage is inferred from topic identity. New sources described here are supplemental and are not counted in the original inventory.
