# Narration and storyboard draft: Enhancer–Promoter Looping as Causal

Authored draft, approximately 60–90 seconds; no rendered video claimed.

Enhancers regulate genes, and bringing a functional looping factor to a silent globin promoter can activate it. But that does not show that enhancer DNA must remain touching promoter DNA throughout every transcriptional burst. Live Sox2 imaging finds enhancer-dependent bursts without a consistent relation to measured proximity. Neural Shh and estrogen-responsive genes add a different warning: activation can increase spatial separation even when ligation contact counts rise. These methods measure different aspects of the nucleus. Acute cohesin loss also leaves most enhancer contacts intact, so it cannot be treated as complete contact removal. Our revised Beta rating applies to the strong continuous-contact necessity claim. It retains causal enhancer and architecture effects while counting contrary native predictions explicitly. Brief earlier encounters remain possible. The decisive next experiment would reversibly release verified contact at a native allele while preserving enhancer chemistry and polymerase competence, then track initiation and active-burst maintenance separately.
