## ADVERSARIAL REVIEW AND APPROVAL

**Authoring model:** Sol 6.1. **Adversarial reviewer:** Claude Opus 5.5 (Anthropic). **Date:** 2026-10-14.

**Scope of checks.** All 60 REFERENCES PMIDs verified against PubMed `get_article_metadata`: 60/60 resolve to the stated first
author, journal, year and DOI; 0 failures; no retraction notice found. Reference 38 is correctly labelled a bioRxiv preprint and
is excluded from primary credit in the REVIEWER NOTES. Twelve load-bearing claims were spot-checked against the cited abstracts
and the selected bodies, concentrating on the ones that hold the global gate shut: the lamin-null embryonic stem cell result that
lamin-B binding correlates with lower expression yet is not required for silencing; the LBR/LAP2 deconvolution's identification of
redundant tethers with reduced H3K27me3 and large long-term deregulation; the B-type lamin speckle study's relocation of
euchromatin away from speckles with global repression, reduced viability and apoptosis; the triple-knockout topology rescue with
matched cell-cycle profiles and no preferential derepression of detached LAD genes; the LAD genetic screen's LMNB1 hit and its
NELF effect shrinking after expression matching; the HDAC1/HDAC3 dissociation of silencing from contact loss; the synthetic 4 Mb
array that transcribes at the lamina; and the Gata4 stage-dependent rescue. Every checked statement matched its source. Scoring
logic was re-derived: caps precede aggregation; the declared `scope-matched` rule is applied as written, fixed to the global SC4
and explicitly refusing to let the narrower Ε-level SC2 set the headline, which is the correct reading of that rule; the Β/Γ/Δ
boundary and the Ζ independence basis were checked. Framework validator re-run.

**Findings**

| ID | Severity | Location | Original to corrected | Evidence |
|---|---|---|---|---|
| N1 | Minor | REVIEWER NOTES | "Consensus browser synthesis will be reconciled ... by the coordinator." to a past-tense sentence pointing at the nine comparisons actually present | The notes promised a reconciliation as future coordinator work while the document already carries a completed "Consensus and original-source reconciliation" section with nine original-source comparisons. The notes contradicted the section they sit above. |
| N2 | Minor | AT A GLANCE, COMPETING MECHANISTIC MODELS, FULL REVIEW, falsification block, REVIEWER NOTES, Consensus section, research timeline and several evidence cards | About eighty glued strings ("The2026LBR/LAP2", "after72h", "laminB1add-back", "chapter18602999,narrative29227210andpreprint38826247were", "showsHDAC1silencing", "H1299CRISPR", "narrowSC2Ε", "a4Mbmulticopytransgene") to spacing restored | Known renderer defect that drops spaces around numerals and between adjacent words. Verified letter by letter against the pre-edit file: no character of content added or removed. |
| N3 | Editorial | Evidence card for PMID16878134, Access/status, and five further Access/status lines | "ErratumIn: PMIDnot supplied." to "Erratum In: notice attached, no PMID supplied."; "ErratumIn:" and "UpdateOf:" to "Erratum In:" and "Update Of:" | Known builder defect producing the glued "PMIDnot supplied" string. The card's own "Does not show" line already records that the linked erratum lacks an identifier, so the corrected wording states the same fact legibly. |
| N4 | Minor | `final_score.confidence_rationale` (scorecard only; not printed in the prose) | Rationale leaning on "incomplete access" to a rationale built on evidence density and reproducibility, with access depth struck | Lead policy: access depth is not evidence density and never justifies a band. Re-deriving from what remains leaves bidirectional expression changes after whole-lamin loss, lamin-null stem-cell competence, a short-versus-chronic detachment discrepancy confounded by stress and death, and unseparated tether redundancy. Those are real reproducibility and confounding reservations, so **Moderate stands**. |

**Rejected findings (checked, no change).**
(i) "Best-supported rival" label: `competing_models.models_evaluated[0]` is the rank-1 model with `is_nominal: false`, so the label
and the printed rank order are both correct. The renderer defect seen in sibling reviews is absent here.
(ii) Scope-matched aggregation: the derivation fixes the headline to the global SC4 and reports the narrower SC2 Ε separately,
which is exactly what the rule requires. No arithmetic or wording error.
(iii) Momentum "Mechanistic only": the artifact names four active mechanistic programmes and states that none completes a
selective global silencer-recruitment proof. Correct value; no "Decisive work underway" claim to test.
(iv) No collective-author defect of either known form. Every first author in the reference list is a personal surname; there is no
leading-comma instance and no organisation fragment such as "Group", "Network" or "Consortium" standing in for a consortium name.
(v) Reference 38 is a 2026 bioRxiv preprint. It is labelled as such in the reference line and excluded from primary causal credit
in the REVIEWER NOTES, and `rests_on_non_peer_reviewed_work` is handled accordingly. Not an error.
(vi) PubMed title fields dropping italicised gene or species names. Declared non-error.
(vii) No spliced "The earlier adversarial approval refers to its original edition" sentence is present.
(viii) The 2026 LBR/LAP2 rival line cites "only minor RNA changes after 72 h detachment" while the PubMed abstract reports
massive deregulation after long-term loss. Checked: the review draws the 72-hour figure from the selected body and explicitly
contrasts short detachment with chronic loss in the Consensus section and in the subclaim rationales. Consistent, not a
misattribution.
(ix) Levels: SC1 Β, SC2 Ε, SC3 Γ, SC4 Γ and the headline Γ follow from the quoted caps and the declared aggregation rule. No
letter required a change.

**Scorecard change.** `final_score.confidence_rationale` only. The level, confidence, subclaim levels, tally and every other
field are unchanged. Validator re-run: PASS.

**Residual limitations.** Eight of 60 records were examined at partial-body depth; the rest are abstract or status examinations,
as the banner states. The citation-context quota was unavailable, as the REVIEWER NOTES record. One erratum remains attached
without an identifier, so its content could not be checked.

**Approval:** Following adversarial review and the corrections listed above, the revised document is approved. - Claude Opus 5.5 (Anthropic), 2026-10-14.
