# Partial Reprogramming and Human Aging

Partial reprogramming has genuine preclinical functional benefits and an early human ocular trial; controlled human healthspan benefit and broad long-term safety remain unestablished.

The preregistered adult-human clinical-healthspan primary remains Γ with Moderate confidence. Direct original-body checks preserve real preclinical function and lifespan benefits while preventing cells, xenografts or remaining mouse lifespan from being presented as treated-human outcomes. A verified first-in-human ER-100 study and sponsor interim signal make this an active translational programme, not an established clinical result. The post-discovery nongating preclinical SC5 receives Ε within its exact experimental contexts.

Species, factor subsets, durations, developmental timing, tissue and clinical readouts show mapped positive/null/hazard boundaries. These support preclinical context-specific SC5 rather than an untested-universal claim. Adult-human clinical healthspan remains a separate unconfirmed scope. Sorting reprogramming intermediates and selecting healthy terminal mouse survivors are real source methods, not eligibility changes that can remove failures from this review.
